Keselowski beat out Erik Jones, Hamlin, Kevin Harvick and Clint Bowyer.
Kyle Busch claimed the regular-season championship while Jimmie Johnson and Alex Bowman secured the final two playoff spots as well.
Ahead of the Brickyard 400 was the Xfinity Series Lilly Diabetes 250, which was won by Justin Allgaier making it his career-best fifth win of the season and his third win in five races.
